The goal of this project is to develop a robust GFP-based imaging technology for studying patterns of retinal cell types and connectivity. This technology will facilitate basic research in the visual system and yield important insights into development, disease, and regeneration of the central nervous system.

Retinal degeneration results in blindness of millions of people. There are over 27 million Americans and 50 million people worldwide currently living with retinal degeneration. The retina is made up of more than 50 cell types organized into six major classes (ganglion, amacrine, bipolar, horizontal, rod, and cone). In human retina, there are more than 20 types of amacrine and ganglion cells, and 100 to 400 types of rod and cone cells. This complexity highlights the importance of studying retinal cell types and their connectivity in order to gain a deeper understanding of retinal development and disease.
The primary limitation of current approaches for detecting different retinal cell types is that retinal cells must be flat-mounted with a small region of interest in one direction. This is necessary to obtain good quality images for many projects. Many projects require the visualization of multiple retinal cell types over a large field of view. Retinal cell types cannot be sorted or analyzed after flat-mounting, so in many cases, imaging is done after clearing the tissue to allow for high-resolution imaging of live-tissue, and cell types can be sorted out or identified after imaging is complete. This requires performing the imaging on the live tissue, which is time consuming and does not lend itself to most investigator’s project plan. This study will develop a robust technology that overcomes these challenges. This project aims to develop and test new tools for multiplex imaging of multiple cell types in an intact, living retina. The tool set will include methods for detecting, imaging, and sorting specific cell types in whole-mount and flat-mounted retinas, and can be used on retinae from zebrafish or other organisms.
